I am in a discussion with my brother regarding the real or autopen issue about an Apollo 11 full crew signature on a Belgian stamp. My father claimed he wrote NASA for this in 1969 and they returned it in a NASA envelope.
How can I tell if the autographs are manually written or not? Thank you.

The signatures are indeed made by an autopen machine. The patterns that the device traced are well documented and can be matched to those on your stamp block.
Still, as a vintage souvenir of the Apollo 11 mission, the block is quite nice!

I lost out to two higher bidders on a very similar item on an eBay auction approximately a year and a half ago. Although the one I bid on was affixed to a cover, and yours has a slightly better placed Armstrong signature but a very inferior placement for the Collins signature.
The cover with mini sheet sold for $75 US. Whether that is average or not I couldn't tell you. Whether the sheet being on cover helps or hinders price wise, I couldn't tell you either.
